在研究体内及体外的免疫系统时,分离淋巴细胞群是一个关键步骤。因为现在市场上并没有针对B细胞的特异性抗血清,所以分离淋巴细胞中的T细胞并不是十分方便。T细胞尼龙毛分离法利用了尼龙毛对B细胞的亲和力,从而使T细胞在没有严重损伤的情况下达到的足够的纯度。因为这个方法不像流式和磁珠费用昂贵,而且操作简便,所需要的条件也不高,是一种非常实用的方法。
首先要指出的是实验中所使用的是分离T细胞专用的尼龙毛(Nylon Fiber),不是化工上的尼龙纤维,曾经有人问过我这种问题,如果你买错了,可是做不出来的!
现在市面上有尼龙毛填料,也已经有了商品化的尼龙毛柱子,这两者的区别主要是以下两点:1.商品化的柱子的填料量是已经定好的,有一个确定的上样量范围。而自己买填料装柱可以控制填料量,也就是可以控制上样量,这对于样本量非常少,或是样本量非常大的实验需求非常合适。2.商品化的柱子已经经过了无菌处理(一般是辐射灭菌),而自己装柱当然就要自己灭菌了。
操作方法:
1.秤取1.5g尼龙毛装入20-30ml玻璃容器或注射器内(要可以灭菌的),填料的体积控制在15毫升左右(注射器上有标记,所以还是比较好控制的),因为尼龙毛的松紧关系到T细胞的回收率,所以要特别注意。注射器下端配接5厘米左右长的带有弹簧夹的橡皮管。
2. 加入大量的PBS平衡柱子后,用铝箔包裹,并置于适当的容器内,高压灭菌15分钟。
(商品化尼龙毛柱以上步骤可以省略,经过PBS平衡后可以直接进行下列步骤。)
3. 灭菌的尼龙毛柱垂直固定后放于超净台内,顶部以铝箔覆盖,避免污染。
4. 注射器下端,橡皮管,弹簧夹用无水酒精消毒。打开弹簧夹调节流速在大约3-4ml/min。
5.首先,加入3-4倍柱体的无血培养基平衡,之后加入相同体积的含有血清的培养基平衡(上述培养基都要预热),最后等培养基液面接近柱填料液面时,关闭弹簧夹。
6. 样本细胞悬浮液浓度控制在约5×108cells/ml,上样量一般是1/3-1/5柱体积。样品加入之后,再加入少量培养液,然后关闭弹簧夹。
7.覆盖铝箔,垂直固定,置于消毒过的容器中,于培养箱内37℃孵育1小时。此过程中柱子必须要保持垂直状态。
8. 从培养箱中拿出柱子,置于超净台内,除去铝箔。接上按照4.中的方法消毒的弹簧夹和橡皮管,加入适当体积经37℃预热的培养基,打开弹簧夹控制流速在3-4 mL/min,流出约5ml之后,流出的培养液基变得不透明,此时其中含有T细胞,收集这一部分培养基。
9. 基本上何时结束收集取决于流出液体中细胞的浓度,实际上,收集的量达到一个柱体积时就可以停止收集,因为即使收集更多的量,回收率几乎不会增加。
10 .上述操作后,实验结果如下:
细胞的回收率为: 15~20 %
T细胞含量: 85~95 %
B细胞污染率: 1 5 %
多数细胞被确定为T细胞。
(注)收集粘附于尼龙毛上的细胞时,将T细胞收集完毕后的尼龙毛再无菌操作的状态下取出,转移到适当的容器中,用镊子小心的将尼龙毛松开,粘附的细胞可以洗到培养基中。或者将注射器芯插入注射器内,通过压力使的粘附的细胞随着液体流出。

实验一 多酚氧化酶(PPO)的分离提取 DEAE-纤维素DE 52

一、原理与目的
多酚氧化酶是植物组织内广泛存在的一种含铜氧化酶,植物受到机械损伤和病菌侵染后,PPO催化酚与O2氧化形成醌,是组织形成褐变,以便损伤恢复,防止或减少感染,提高抗病能力。醌类物质对微生物有毒害作用,所以伤口醌类物质出现是植物防止伤口感染的愈伤反应,因而受伤组织一般这种酶的活性就会提高。多酚氧化酶也可与细胞内其他底物氧化相偶联,起到末端氧化酶的作用。
PPO的存在是水果、蔬菜褐变及营养丧失的主要原因之一。PPO氧化内源的酚类物质生成邻醌,邻醌再相互聚合成醌或蛋白质、氨基酸等作用生成高分子络合物而导致褐色素的生成,色素分子量愈高,颜色愈暗。多酚氧化酶活性高低也是马铃薯解除休眠的指标之一。
本实验将采用马铃薯为主要材料,通过组织细胞破碎匀浆、过滤、离心、硫酸铵沉淀、透析等步骤获得PPO的粗酶液。
通过本项实验,学习和了解蛋白质的提取、分离的基本原理和方法,掌握相关仪器设备的操作使用,以及蛋白质的提取、分离的系统技术。
二、材料与试剂
(1)马铃薯(大约每小组100-200g)
(2)试剂:
0.03M磷酸缓冲液PH6.0(内含0.02M巯基乙醇,0.001MEDTA,5%甘油,1%的聚乙烯吡咯烷酮)配制时配×10倍的浓缩液1000ml;固体硫酸铵;0.03M磷酸缓冲液PH6.0(内含0.02M巯基乙醇,0.001MEDTA,0.005MgCL2)配置时配
(3)实验器械与仪器设备:
试管与试管架;烧杯、玻璃搅棒;移液管、滴管等;试剂瓶;透析袋;
过滤纱布;植物组织匀浆器;pH计和pH试纸;GL-20C高速冷冻离心机;
DL-7A大容量低速冷冻离心机
三、操作步骤
1:粗酶提取:
水果肉组织按1:1(W/V)比例与003M磷酸缓冲液PH6.0(内含0.02M巯基乙醇,0.001MEDTA,5%甘油,1%聚乙烯吡咯烷酮)混合,匀浆4层后纱布过滤,滤液以6000rpm离心10min,弃除沉淀获得酶提取液。
2:盐析分级沉淀:
在酶提取液中加入固体硫酸铵使其达到40%饱和度(边加边搅拌达到充分溶解),然后6000rpm离心20min弃除沉淀;离心上清液在加入固体硫酸铵达到70%饱和度(边加边搅拌达到充分溶解),再以6000rpm离心20min,弃除上清夜,收集粗酶沉淀。
四、实验结果
获得PPO粗酶液。
实验二 PPO粗酶液的纯化
一、原理
1.葡聚糖凝胶Sephadex G-200凝胶柱层析 利用大分子不能进入凝胶颗粒内部 最先流出柱外,而小分子物质进入凝胶颗粒内部,流速慢而最后流出柱外,凝胶层析法能将不同分子大小的物质分离开。(G-200能分离800-80000D分子量的蛋白质)
2.利用离子交换法,选取DEAE-阴离子纤维素DE52柱材料,因为PPO是带有阳离子的蛋白质,在层析时会吸附在柱材料上,而杂蛋白会洗下。后用NaCl梯度洗脱PPO,(NaCl为强离子交换剂,能将弱吸附在柱材料上的PPO置换下来)。粗酶液从而得到纯化。
二、材料与试剂
试剂:0.02M Tris-HCL缓冲液Ph7.4(内含0.001M EDTA)配制时配×10倍浓缩液1000ml;NaCL;聚已二醇;DEAE-纤维素DE52;葡聚糖凝胶Sephadex G-200;兰葡萄糖-40、70、100等;
实验器械与仪器设备:试管与试管架、烧杯、玻璃搅棒、移液管、滴管等;试剂瓶、层析柱、pH计和pH试纸、恒流泵、梯度混合仪、核酸蛋白监测仪、GL-20C高速冷冻离心机、DL-7A大容量低速冷冻离心机
三、操作步骤
1.葡聚糖凝胶的处理、装柱及平衡
(1)柱材处理
称取一定量的Sephadex G-200干粉,加无离子水或蒸馏水浸泡溶胀48h,去掉悬浮的细微颗粒,为防止凝胶颗粒中的空气存在,影响层析效果,凝胶装柱前一定要将凝胶液中的气体除掉,其办法是将凝胶液放进抽滤瓶中,进行抽真空处理,直到凝胶液中没有气泡出现为止。
(2)装柱
将层析柱清洗干净垂直固定到层析架上,加1/3体积的无离子水,打开下出液口,水流畅通,即刻将小烧杯中装有适宜浓度的凝胶液(凝胶的浓度过高会产生气泡,影响蛋白质分离速度,浓度过低易产生凝胶分层,影响蛋白质的分离效果)轻轻倒入层析柱中,凝胶自然慢慢沉降在层析柱底部,凝胶沉积直到离层析柱上端1.5-2cm处,停止装柱,剪一与柱内径相等的滤纸片覆盖于凝胶上。层析柱上端进液口连接恒流泵,下出液口连接蛋白质监测仪,待层析柱的平衡。
(3)平衡
在柱层析上样前必须对层析柱进行平衡,所谓平衡就是将层析柱中的溶液用层析过程的缓冲液(洗脱液)置换出来,使层析柱中的缓冲系统与柱层析过程中的系统一致。其方法是:利用层析柱上端的恒流泵将平衡缓冲液泵入到层析柱内,打开层析柱下端的出口,平衡液流速在0.5-1ml/min,当下出口流出液的PH值与平衡缓冲液的PH值一致时,层析柱达到了平衡。在本实验中,用0.002M Tris-HCL缓冲液PH7.4(内含0.0001M EDTA),平衡Sephadex G-200凝胶。
2.上样:将粗酶液上柱。
3.洗脱:用0.02M Tris-HCL缓冲液Ph7.4(内含0.001M EDTA)洗脱,收集洗脱液进行酶活性、蛋白质浓度、聚丙烯酰胺凝胶电泳分子量的基本性质分析测定。
4.DEAE-纤维素的处理及装柱
(1)处理
本实验采用的是DEAE-纤维素DE 52 是弱酸型阴离子交换剂,具体处理方法为:先将DE52阴离子交换剂干粉浸泡于蒸馏水中,去除杂质;再在0.5N的HCL溶液中浸泡1-2h,再用无离子水或蒸馏水洗至pH值中性或PH4以上,并将其在抽滤漏斗中抽干;将抽干的离子交换剂浸泡在0.5N的NaOH溶液中1-2h,再用无离子水或蒸馏水将其洗至中性。
(2)装柱
将层析柱清洗干净垂直固定到层析架上,加1/3体积的无离子水,打开下出液口,水流畅通,即刻将小烧杯中装有适宜浓度的柱材,轻轻倒入层析柱中,凝胶自然慢慢沉降再层析柱底部,凝胶沉积直到离层析柱上端1.5-2cm处,停止装柱。层析柱上端进液口连接恒流泵,下出口连接蛋白质监测仪,待层析柱的平衡。
(3)平衡

5.上样:
将洗脱酶液上样,用0.02M Tris-HCL缓冲液pH7.4(内含0.001M EDTA)洗脱杂蛋白。
6.洗脱:
用含NaCL(0.2-0.6M)的0.02M Tris-HCL缓冲液pH7.4(内含0.001M EDTA)进行梯度洗脱。
7.测定酶活性和蛋白质浓度
实验三 PPO活性测定
一、原理与方法
PPO催化各种酚与O2氧化为醌。本实验是采用邻苯二酚为底物,在0.2M磷酸氢二钠-0.1M柠檬酸缓冲液PH5.8的反应体系中,PPO催化邻苯二酚形成褐色的醌,在分光光度计410nm处使反应体系的OD值产生变化,通过OD值上升的读数变化确定PPO的酶活大小。
其方法是:在含有4mlPH5.8的0.2M磷酸氢二钠-0.1M柠檬酸缓冲液试管中加入0.05ml0.05M邻苯二酚溶液,在30℃恒温水浴中预热后加入0.05ml酶液,反应5分钟,在分光光度计410nm处读取吸光值。在上述条件下,以A410读数,以每分钟增加0.01O.D.值定义为一个酶活性单位(U)。

分析:酶活性高低不一定与蛋白含量高低成正比,3和4试管中含较多所需蛋白。
实验四 胰酶分离提取
一、原理与目的
提取制备酶的经典方法,多采用硫酸铵分级沉淀,胰酶在75%饱和度硫酸铵中沉淀,其它杂蛋白一般在10%硫酸铵饱和度下被沉淀而除去。经此多次提取,最后在PH8.0硼酸缓冲液中得到结晶。
胰酶在PH3.0时最稳定,可在低温下储存较长的时间而不失活;低与此PH酶易变性;高于PH5时,酶易自溶而失活。因此提取制备胰酶的全部操作过程应在低温和低PH下进行。胰酶以无活性的酶原形式存在于动物的胰脏中。胰蛋白酶原在正常生理条件下可被肠激酶和钙离子所激活或自我环化成为有活性的酶。猪胰蛋白酶原分子量约24000-25000,而猪胰酶分子量为23400。在酶原活化的过程中,酶原N端Lys与Ile之间的 一个肽链被断裂,丢失一个6肽,分子构象发生改变,PI变成10.8。
本实验拟通过从猪胰脏中制备胰酶结晶实验,掌握酶的制备、一般分离、提取、纯化和结晶的操作技术。同时为亲和层析、免疫学实验准备实验样品。
二、材料与试剂
1.仪器
紫外光分光光度计、恒温水浴、离心机、组织捣碎机、PH计、显微镜、秒表、剪刀、镊子、搪瓷盆、乳钵、大玻璃漏斗、抽滤瓶,塑料小桶、纱布、PH试纸,滤纸、玻璃器皿等。
2.材料:新鲜猪胰脏
3.试剂及溶液配制
pH2.5乙酸酸化水、2.5M H2SO4溶液、2M.NaOH溶液、5M NaOH溶液、01M HCL溶液、0.025M HCL溶液、0.01M HCL溶液、0.4M pH9.0硼酸缓冲液(母液为0.8M,用时稀释一倍)、Tris、硫酸铵。
0.8M pH9.0硼酸缓冲液:取20ml0.8M四硼酸钠溶液,混合后用PH计校正。
三、操作步骤
1.胰蛋白酶原的提取与分离:
(1)称取1-1.5Kg新鲜猪胰脏,在冰浴下剥除脂肪和结缔组织,在低温下用绞肉机绞碎胰脏(或用高速组织匀浆机捣碎)加1-2倍体积以预冷却的PH2.5-3.0乙酸酸化水溶液提取(按1g组织相当于1ml体积计算,以此类推)。检查提取液中PH,若高于PH3.0时应及时用10%乙酸调节,使提取液维持PH2.5-3.0之间。在冷室5℃左右搅拌提取18-24h,而后用4层纱布过滤,尽量拧挤出滤液,滤液呈乳白色,用约300mlpH2.5乙酸酸化水再提取残渣一次(时间约为1-2h),合并滤液,此时滤液PH值较高,可用5M H2PO4溶液调整PH至2.5-3.0,放置 3-5h后,浑浊滤液冷却后,用玻璃漏斗进行自然过滤,滤液呈黄色透明液体,收集滤液,量其总体积,弃去沉淀物。
(2)盐析:滤液加固体粉状硫酸铵进行盐析,使溶液至75%饱和度。盐析溶液放冷室中过夜,次日用4000r/min离心机离心,或用布氏漏斗抽滤,滤饼经压干后称重,可得约20g胰蛋白酶原粗制品。
(3)胰蛋白酶原得激活:将胰蛋白酶原粗制品用10倍体积得冷蒸馏水,分多次加入使滤饼溶解,溶液呈乳白色,量其体积,取出1ml溶液用于测定,溶液中硫酸铵得含量(约为滤饼重得1/4)。因为硫酸铵可以与活化剂钙离子结合,生成硫酸钙,如果钙离子过少会直接影响胰酶原的激活过程,按浓度量计算,将已研细的固体CaCL2慢慢加入酶原溶液中,边加边搅拌均匀。用5MNaOH溶液调PH至8.0。在酶溶液加入约5mg结晶猪胰蛋白酶,轻轻搅拌均匀,置5℃冰箱中使酶原活化。
(4)纯化:去除杂蛋白,量取胰蛋白酶溶液体积后,用5M硫酸溶液调节滤液PH至2.5-3.0,抽滤除去硫酸钙沉淀,滤液体积约1000ml,加入已经研细的硫酸铵粉末,使其溶液达40%饱和度(每升滤液加242g硫酸铵)。放置5℃冰箱中5-8h,抽滤除去沉淀。
实验五 卵清蛋白分离提取
一、实验目的与原理
鸡卵粘蛋白存在于鸡蛋清中,对胰蛋白酶有强烈的抑制作用,高纯度的鸡卵粘蛋白抑制胰蛋白酶的分子酶的分子比为1:1。鸡卵粘蛋白在中性或酸性溶液中对热和高浓度的脲都是相当稳定的,而在碱性溶液中较不稳定。
由于鸡卵粘蛋白对胰蛋白酶有强烈的抑制作用,因此可以用鸡卵粘蛋白做亲和配基配制纯化胰酶的亲和材料。
二、材料与试剂:
1.材料:新鲜鸡蛋2只
2:仪器:抽滤瓶500-1000ml、烧结漏斗、移液器、磁力搅拌器
3:试剂:
10%TCA,用固体NaOH调调PH至1.05-1.10,需要50ml、5N HCL、5N NaOH、冷丙酮、胰蛋白酶液、BAEE-0.05M,PH8.0Tris-HCL缓冲液(每ml含0.34BAEE和2.22mgCaCL2),50ml
pH8.0,0.1M Tris-HCL缓冲液
三、操作步骤
1,取两只新鲜鸡蛋,得蛋清50ml,置于烧杯中,外用温水浴25℃-30℃,在不断搅拌条件下,缓慢加入等体积得三氯乙酸-丙酮(1:2V/V),立即出现大量白色絮状沉淀,加完后最终PH约3.5,再继续搅拌30min,然后在4℃冰箱中放置过夜。
2,次日用布氏漏斗抽滤,得黄绿色清液。
3,边搅拌边加入4℃预冷的丙酮200ml沉淀蛋白,在4℃放置2h之后将上清液小心倒入瓶中回收,下部沉淀部分于4000rpm离心5min,收集沉淀。
4,将沉淀溶于10ml无离子水中,对无离子水(50倍)透析4h,换水两次,再对碳酸钠缓冲液透析过夜,4000rpm离心10min ,去除不溶物。
实验六 亲和层析纯化胰酶
一、实验目的与原理
生物大分子化合物的重要特征之一是具有与某些相对应的分子通过次级键或共价键专一结合的能力即亲和力。例如酶与抑制剂之间,抗原与抗体之间,结合的双方不仅是专一的,而且结合以后可以在不丧失生物活性的基础上用物理或化学的方法进行解离。根据这一特性,如果把具有亲和力的一对分子的某一方连接于水不溶的固体载体上作为固定相,那么另一方随着流动相流经该固定相的时候,双方便亲和结合为一个整体。然后只有改变某种物理条件或化学条件使结合的双方解离,即能得到于固定相有特异亲和力的某一特定物质。这种利用生物分子和相对应分子之间亲和结合和解离性质而建立起来的层析方法称之为亲和层析。
本实验通过将胰蛋白酶抑制剂(protease inhibitor)(protease inhibitor)(protease inhibitor)-鸡卵粘蛋白与Sephadex-G75偶联及亲和层析纯化胰酶的操作,以了解亲和层析的基本原理,掌握亲和柱材活化偶联及亲和层析操作的基本过程和技术。
二、仪器与试剂
1.鸡卵粘蛋白制备
2.Sephadex-G75的活化和偶联
2.5g干重的葡聚糖凝胶Sephadex-G75 、0.05M NaCL溶液 50ml、纯化的鸡卵粘蛋白87.5mg、 5% K2CO3、0.27gKBH4 (溶于20ml水)
3.亲和层析
0.1M Tris-HCL pH7.5(含0.5M KCL,0.05M CaCL2) 200ml、粗胰蛋白酶约50ml 、0.1N甲酸钾、0.5M KCL pH2.5 100ml
仪器器材:抽滤瓶500-1000ml、层析柱、紫外分光光度计、紫外蛋白检测仪、烧杯漏斗、移液管、磁力搅拌器
三、操作步骤
1、鸡卵粘蛋白的制备
2、Sephadex-G75的活化及偶联
2.5g干重的葡聚糖凝胶Sephadex-G75溶胀洗涤数次,缓慢搅拌,加入0.05M NaCL溶液50ml ,30min,蒸馏水洗涤数次,抽干备用,活化凝胶,纯化的鸡卵粘蛋白87.5mg 溶于35ml水,加入活化葡聚糖凝胶,75%K2CO3,调pH7.5-9.0,缓慢搅拌3h。反应过程随时加入5%K2CO3,以维持pH的稳定。0.27gKBH4 溶于20ml水,缓慢搅拌加入上述体系反应5h,pH维持在6.5-7.5,洗涤。
3、胰蛋白酶的亲和层析
卵类粘蛋白在pH7-8的条件下能与胰蛋白酶专一地结合,而在pH2-3时又能解离,因而掌握好上柱地PH条件和洗脱缓冲条件,便可将胰蛋白酶从含杂蛋白地粗酶液中选择性地结合于柱上,待洗去不结合或非专一性结合地杂蛋白后,改变PH值便可将胰蛋白酶洗脱下来。从而达到纯化地目的。由于亲和层析结合地专一性,上柱体积可以比较大,得到浓缩的提纯产品。
(1)亲和层析
将鸡卵粘蛋白-Sephadex-G75装柱(1×10cm),用pH7.5,0.1M含 0.5M KCL,0.05M CaCL2的缓冲液平衡,平衡约2-3倍的柱体积。将粗胰酶液上柱层析。用紫外蛋白监测仪280nm处检测蛋白质吸收峰,随着上柱和流出的杂蛋白量的平衡,吸收峰达到稳定,一旦上柱的胰蛋白酶量超过柱的负荷时,则胰蛋白酶溢出,使吸收峰升高,此时应停止样品上样,改用pH7.5的平衡缓冲液冲洗出杂蛋白,待洗出液的吸收回到基线后,改用0.1N的甲酸钾0.5M KCL,Ph2.5的洗脱液解析,柱的流速维持在1.5ml/min左右,收集洗脱下来的蛋白峰蛋白,测总蛋白量及比活性,并根据上柱样品的总蛋白量和比活性计算经亲和层析后胰蛋白酶比活性提高的倍数,总蛋白回收率,胰蛋白酶量。
当胰蛋白酶峰洗出,待紫外吸收回到基线后,重新用缓冲液平衡,亲和层析柱可以反复使用。

Whatman Anopore 无机膜是各种实验室过滤的理想用品。这种新颖的材质制成的膜具有精
确、不变形的蜂窝形孔结构,并且每个孔之间没有横向交叉,这种特性确保了小于孔径的
颗粒滤过,而大于孔径的颗粒被阻截。Anopore 无机膜由高纯度的氧化铝基质通过电化学
制成,蛋白吸附力低、自身荧光非常小、无毒、支持细胞生长。
精确的孔结构和均匀的孔分布,无机膜能确保高效滤除颗粒。微生物和颗粒物保留在膜表
面,便于进一步用电子显微镜分析。经湿润后,膜变成半透明,因而不需对截留物进行转
移就可用光学显微镜观察。
无机膜是亲水性的,能与大多数溶剂和水溶液相容。因生产过程中不另加单体、高分子物
质、表面活性剂或润湿剂避免了样品污染。低蛋白吸附力,可减少样品的损失。
无机膜以Anodisc形式供应,也就是在无机膜边缘加上一个聚丙烯环(除13mm规格圆片)
,便于操作,适用于真空和压力过滤。
Anopore有三种孔径:0.02μm、0.1μm和0.2μm;和三种直径:13mm、25mm和47mm。

whatman 定量滤纸——硬化低灰级

GE Whatman-Xinhua Filter Paper 沃华滤纸
这些滤纸的最高灰分含量介于无灰级和定性级之间。非常合适用于布氏漏斗过滤。其坚韧光滑的表面易于回收沉淀物。其他特性包括高湿强以及化学相容性都类似于硬化无灰级别。
Grade 50:2.7µm
可以保留非常细小的结晶物沉淀。是whatman滤纸中最薄的一种。流速慢,颗粒保留性好。硬化的表面有效地防止纤维脱落。非常合适用于布氏漏斗、三件套漏斗真空定性或者定量过滤。很好的湿强,可以在湿润状态下处理沉淀刮除。

Whatman专利产品FTA卡是在室温下对各种生物样品进行收集、运输、存档和纯化其核酸并进行PCR、SNP分析、RT-PCR、RFLP以及限制酶解作用分析而设计的,可应用于任何样品如血液、微生物、细菌、植物物质、口腔细胞、固体组织、培养的细胞、病毒、M13斑及其它样品。基因DNA在FTA卡上可在室温下储存11年以上而不会减少PCR效率。FTA卡经专利的化学配方浸渍,能溶解细胞膜并改变蛋白质特性。核酸被固定下来并可保护其免于UV光线损害、以及微生物和真菌的侵蚀,样品中的传染性病原体在FTA卡上也失去活性,样品收集在FTA卡上非常安全,可通过邮局寄送不用贴危险标记。要回收已捕获的核酸,需从FTA卡上取下一小圆片进行洗脱,让洁净的核酸留在小圆片上,然后可将小圆片直接用于下游的应用,如PCR等(即使经过PCR反应后,模板DNA仍然吸附在FTA小圆片上,因而可重复做PCR及其它分析)。清透的样品建议用指示型FTA卡,指示型FTA卡在样品加入后会变色(粉红变成白色),便于辨认样品的位置。质粒DNA的提取请用CloneSaverTM卡。FTA标准卡有4个收集片,通用于各项的应用:FTA基因卡有3个收集片,适用于亲子鉴定;FTA微型卡有1个收集片,适用于样品量少的应用。

MAXYGEN  PCR仪采用了独特的样品温度和模块温度同步技术,保证实验结果的重现性。在实验中我们发现样品的温度变化最快在1℃左右,模块温度过快变化导致样品温度滞后于模块温度,导致了实验条件的失真。MAXYGEN PCR仪采用了2-3度的变化速率,到临近设定目标温度时减速,该智能控制模式使样品和模块温度保持高度一致,最大的保证了实验结果的重现性和条件的真实性;到达预订温度后如何使温度稳定而不出现波动的难题一直困扰着许多厂家,MAXYGEN PCR仪采用的温控系统具备了极高的稳定性,消除了在到达预设温度后出现的上下波动。
MAXYGEN  PCR仪的出现让你告别了过去命令行式的原始操作,超大的LCD面板,生动直观的图形化程序编制让初学者一看就会,节省使用者大量的学习时间。系统支持模块化插入程序段,预设有标准程序段,利用该标准程序段,轻轻移动光标就可以编制出复杂的温度控制程序,永别一行一行设定参数的日子!
MAXYGEN  PCR仪采用了滑盖设计,有效的避免了传统翻盖开盖时高温烫伤现象。在热盖温度上,有60—65℃和100-115℃两个范围可选,在PCR管密封性不太好的情况下,可以采用提高热盖温度的方法防止蒸发,而固定温度无法做到这点。另外热盖还可以设定在60-65度,用于酶切等反应。同时热盖高度可调,使用不同品牌不同规格PCR管,到达一定压力会有提示音,提示管盖压力已到位,避免用力过大损坏PCR管或者压力过小导致密封性不够!

7000型荧光定量PCR仪的多种应用
1.绝对定量 通过测量样本的Ct值和采用标准曲线,可定量分析未知样品的起始拷贝数。标准曲线是通过计算稀释的已知拷贝数样本的Ct值而得出的。
2.相对定量 相对定量分析通过一个标准样本来计算表达水平,而不是使用标准曲线。使用内源性的对照来校正样品的加样量。
3.高分辨的SNP检测 7000型荧光定量PCR仪还能利用多色荧光检测单核苷酸多态性(SNPs)。
3.阴/阳性分析(终点分析) 使用内部阳性对照(IPC),7000型荧光定量PCR仪可进行阴/阳性分析。由于每个待检测样品中都加入阳性对照品,可以有效防止假阴性的结果。
