CHT 陶瓷羟磷灰石 (Ca5(PO4)3OH)2 介质是当今要求极高的下游过程工艺中主流的纯化介质。这种混合介质具有独特的选择性和分辨率,经常能分离一些其它层析和电泳技术无法区分的蛋白质。CHT介质能够结合宿主细胞蛋白质、滤取的 A 蛋白、抗体调光器和集合能够用在从样品捕获到精纯的每一步。
CHT 陶瓷羟磷灰石具有以下稳定的特性,因而可提高效率、产量和经济价值:
- 在高流速下捕获效果优异,可应用于小试研究到规模放大

- 优异的选择性,可以进行两步式色谱处理
羟磷灰石 (Ca5(PO4)3OH)2 是磷酸钙的一种形式,可用于生物分子的层析分离。 五个钙双联体(C 位点)和一对含羟基的磷三联体(P 位点)组按一个重复的几何图形排列。 在该材料的电子显微照片中可看到重复六边形结构;其拉曼光谱中的空间填充模型和重复结构目前也已完成。
两种类型的 CHT 陶瓷羟磷灰石(I 型和 II 型)介质均有 20、40 和 80 µm 三种大小不同的颗粒尺度。两个类型介质的洗脱特性都具有类似于晶体羟基磷灰石,但还有一些重要差别:
- I 型,在 400°C 下烧结,蛋白载量高,且对酸性蛋白载量更高
- II 型,在 700°C 下烧结,蛋白载量较低,对早期洗脱蛋白及核酸的分辨率较佳,适合于纯化多种类的免疫球蛋白
