描述

HRP、AP 等酶可把某些底物转变为有色沉淀。当沉淀在印迹膜上聚集，就产生了可见的有色信号。反应可监控，当观察到目标信号超过背景时，即停止反应。比色检测比以胶片为基础的化学发光检测操作更简单; 然而，此方法的单一的最终结果不能产生化学发光法所拥有的多次曝光能力。与放射性或化学发光检测相比，一般认为比色检测是一种中度灵敏的方法。

Bio-Rad 基于 AP 的比色试剂盒利用 5-溴-4-氯-3-吲哚基磷酸酯/硝基蓝四氮唑 (BCIP/NBT)，这种物质可与碱性磷酸酶发生反应形成有色沉淀以便检测。 Immun-Blot AP 测定试剂盒包括比色检测所必需的所有试剂，最多可检测 100 pg 的蛋白质。 为获得较高灵敏度，Immun-Blot 扩增型 AP 测定试剂盒使用生物素-链亲和素共轭系统来扩增比色信号，可以检测低至 10 pg 的蛋白质。
